چکیده
Abstract Soil salinity severely hampers agricultural productivity. Under salt stress, excess Na + accumulation causes cellular damage and plant growth retardation, membrane transporters play central roles in uptake exclusion to mitigate these adverse effects. In this study, we performed sos1 suppressor mutant (named sup ) screening uncover potential genetic interactors of SOS1 additional tolerance mechanisms. Map-based cloning sequencing identified a group mutants harboring dominant gain-of-function mutations the vacuolar /H antiporter gene AtNHX1 . The variants showed enhanced transporter activities yeast cells increased Arabidopsis wild type plants. Ion content measurements indicated that at level, resulted likely due sequestration. However, reduced shoot but root under indicating role limiting translocation from shoot. We also another suppressors with loss-of-function AtHKT1 Loss-of-function additively suppressed sensitivity, which indicates three transporters, SOS1, function independently coordinately controlling homeostasis Arabidopsis. Our findings provide valuable information about target amino acids NHX1 for editing improve crops.

Improved salt tolerance in canola (Brasica napus) plants by overexpression of Arabidopsis Na+/H+ antiporter gene AtNHX1
A significant portion of the world’s cultivated land is affected by salinity that reduces crop productivity in these areas. Breeding for salt tolerance is one of the important strategies to overcome this problem. Recently, genetic engineering is becoming a promising approach to improving salt tolerance. In order to improve the yield performance of canola in saline soils, we transformed canola w...
متن کاملArabidopsis sos1 mutant in a salt-tolerant accession revealed an importance of salt acclimation ability in plant salt tolerance
An analysis of the salinity tolerance of 354 Arabidopsis thaliana accessions showed that some accessions were more tolerant to salt shock than the reference accession, Col-0, when transferred from 0 to 225 mM NaCl. In addition, several accessions, including Zu-0, showed marked acquired salt tolerance after exposure to moderate salt stress. It is likely therefore that Arabidopsis plants have at ...
متن کاملOverexpression of SOS (Salt Overly Sensitive) genes increases salt tolerance in transgenic Arabidopsis.
Soil salinity is a major abiotic stress that decreases plant growth and productivity. Recently, it was reported that plants overexpressing AtNHX1 or SOS1 have significantly increased salt tolerance. To test whether overexpression of multiple genes can improve plant salt tolerance even more, we produced six different transgenic Arabidopsis plants that overexpress AtNHX1, SOS3, AtNHX1+SOS3, SOS1,...
متن کاملCo-overexpressing a Plasma Membrane and a Vacuolar Membrane Sodium/Proton Antiporter Significantly Improves Salt Tolerance in Transgenic Arabidopsis Plants
The Arabidopsis gene AtNHX1 encodes a vacuolar membrane-bound sodium/proton (Na(+)/H(+)) antiporter that transports Na(+) into the vacuole and exports H(+) into the cytoplasm. The Arabidopsis gene SOS1 encodes a plasma membrane-bound Na(+)/H(+) antiporter that exports Na(+) to the extracellular space and imports H(+) into the plant cell. Plants rely on these enzymes either to keep Na(+) out of ...
متن کاملComparative genomics in salt tolerance between Arabidopsis and aRabidopsis-related halophyte salt cress using Arabidopsis microarray.
Salt cress (Thellungiella halophila), a halophyte, is a genetic model system with a small plant size, short life cycle, copious seed production, small genome size, and an efficient transformation. Its genes have a high sequence identity (90%-95% at cDNA level) to genes of its close relative, Arabidopsis. These qualities are advantageous not only in genetics but also in genomics, such as gene ex...
